Uprima Information
Common Names: Uprima, Uprime
Uprima is a prescription medication used for the treatment of erectile dysfunction or ED. Erectile dysfunction is a condition in which a man cannot achieve an erection sufficient enough for intercourse. Uprima helps men achieve erections by stimulating the region of the brain called the hypothalamus. As a result of a dose of Uprima, the body increases the production of dopamine, an important chemical in the body that helps trigger erection.
Uprima Ingredients and Composition
Each Uprima tablet contains 2 or 3 mg apomorphine HCl hemihydrate.
An Uprima tablet also contains the following non-active excipients:
- microcrystalline cellulose
- hydroxypropyl
- methylcellulose
- citric acid
- magnesium stearate
- ascorbic acid
- edetate disodium dihydrate
- colloidal silicon dioxide
- ferric oxide
- acesulfame potassium
- orange mint flavour and a sufficient amount of mannitol to attain the final tablet weight.
There are no additional Uprima ingredients.
How Does Uprima Work?
Uprima (apomorphine) is a dopaminergic agonist with affinity for both D1 and D2 receptors in sites within the brain known to be involved in the mediation of erection. Uprima studies in vivo have shown the erectile function effects of apomorphine are mediated at dopamine receptors in various nuclei in the hypothalamus and midbrain. In particular, the paraventricular nucleus of the hypothalamus has been identified as the site of action. This site may mediate autonomic aspects of sexual arousal. Oxytocinergic and nitric oxide signalling may be involved in the cascade of neural events that result from the central action of apomorphine.
Apomorphine acts as a central initiator of erection and enhances pro-erectile stimuli. The erectogenic effects of Uprima arise from improved central neural signalling specific to penile vascular response.
In simple terms:
Uprima, unlike other oral treatments for erectile dysfunction, stimulates dopamine receptors in the brain. These receptors in turn transmit excitatory signals down the spinal cord to the sacral parasympathetic nucleus, stimulating activity of the sacral nerves supplying the penis. As the penile smooth muscle relaxes, the pelvic arteries dilate. The dilated arteries then fill with blood and the penis becomes erect.
How To Take Uprima and Uprima Dosage and Administration
It is recommended that you take the Uprima sublingual tablet 20 minutes prior to sexual activity. It is best to drink a small glass of water before taking the Uprima tablet to help the dissolving process.
After drinking the water, place the Uprima tablet under the tongue. Do not chew, just let the tablet dissolve. In the majority of patients, the Uprima tablet will be dissolved within 10 minutes. An erection should occur when sexual activity starts.
It might take about 10 minutes for the Uprima tablet to dissolve. Sexual stimulation is required for the drug to take effect, and for an erection to be achieved. For most patients, Uprima should be taken once a day as required. Take one Uprima tablet in an eight-hour period. Do not exceed 2 mg within an 8 hour period, as serious complications can occur.
If you suspect a Uprima Overdose
Uprima may incude vomiting if taken in high doses. If Uprima tablets are swallowed, the absorption of apomorphine will be reduced by first-pass hepatic metabolism. There is no specific antidote available for Uprima. Treatment should be supportive and symptomatic. It is advised that vital signs, such as blood pressure and heart rate, be monitored. Measures to avoid possible orthostatic hypotension should be taken. The use of domperidone maleate, a peripherally acting dopamine antagonist used to counter emetic effects, may be considered in case of an Uprima overdose.
Uprima Side Effects
As with other errectile dysfunction treatments, you may experience side effects with Uprima. Uprima side effects are generally mild in nature. In most cases they do not require you to quit treatment as a result. Common Uprima side effects include:
- nausea
- headache
- dizziness
- yawning
- sleepiness
- infection
- sore throat
- pain
- increased cough
- rhinitis
- flushing
- taste disorder
- sweating
In very rare incidences fainting might occur as an Uprima side effect. If you feel faint, lie down and raise your legs. This should help alleviate any symptoms of faintness. If you experience chest pains during intercourse, stop immediately and contact your doctor. If prolonged Uprima side effects occur or if you experience any one of the previous symptoms, stop treatment and consult with your doctor immediately.
There may be additional Uprima side effects.
Uprima Precautions and Contraindications
Uprima is a prescription medication intended for use in generally healthy male patients. It is not approved for women or children.
Do not administer Uprima if:
- You have ever had an allergic reaction to apomorphine or any other ingredients found in Uprima.
- You suffer from severe unstable angina, recent myocardial infarction, severe heart failure, hypertension, or any other heart problems.
- Your partner is pregnant or breast-feeding.
If you fall under any one of the following conditions, use extra precautions and consult your doctor before taking Uprima:
- You have a deformity or disease of the penis.
- You experience severe dizziness or feel faint after taking Uprima.
- You are taking medications with nitrates.
- You have liver or kidney problems.
- You have suffer from hypertension (high blood pressure).
- You are taking other treatments for erectile dysfunction.
Uprima Clinical Trials and Studies
In clinical studies, Uprima was administered to men with organic, psychogenic, or mixed etiology erectile dysfunction, and was evaluated for its ability to produce an erection firm enough for intercourse. Initial Uprima clinical studies included patients with controlled hypertension and diabetes (type I and type II). TAP has also evaluated Uprima in patients following nerve-sparing radical prostatectomy.
In addition, a unique aspect of TAP's Uprima clinical studies was the assessment of patients' partners for satisfaction. This assessment showed corroboration of findings from the patient.
The most commonly reported Uprima side effect was nausea. Of the nausea reported in the NDA clinical studies, most incidences were mild to moderate in severity.
The manufacturer of Uprima makes the following claim, which has not been substantiated by The Pharma-Help Team:
Once Uprima is dissolved under the tongue, clinical test have shown that 34% of erections occurred within 10 minutes and 71% of erections occurred within 20 minutes. That puts the average time to the onset of an Uprima erection at 18-19 minutes - 200% faster than a leading competitor brand.
Storing Uprima
Keep Uprima out of reach and sight of children. Store Uprima in the original package. Do not use Uprima after the expiration date shown, or if the pack has been damaged or tampered with.
